Intralysosomal cystine crystals in cystinosis.

چکیده
Electron microscopic study of conjunctival biopsies from members of one sibship each of nephropathic and benign cystinosis showed polymorphic cystine profiles abundantly distributed in fibroblasts and histiocytes. These crystalline spaces measured from 0.2(i to 2/i and were delineated marginally by intact cellular membranes. Small crystalline aggregates were found in acid-phosphatase-positive osmophilic dense bodies and they appear to be the earliest manifestation of intracellular deposition of cystine. This study suggests that there are striking morphologic similarities in the tioo forms of cystinosis at the cellular level and. supports the suggestion that both may represent disorders in cystine metabolism by a subcellular organelle of lysosomal type.

Introduction Nephropathic cystinosis is a rare autosomal recessive disorder caused by a mutation in the CTNS gene, which encodes for cystinosin. It is characterized by the lysosomal accumulation of cystine, which leads to the formation of cystine crystals within various organs, including kidneys, brain, cornea, intestine and bone marrow. The exact role of intralysosomal cystine crystals accumul...
متن کاملDisulphide reduction in lysosomes. The role of cysteine.
Published evidence indicates that cystine-containing proteins have their disulphide bonds reduced during proteolysis in lysosomes. However, the intralysosomal accumulation of cystine in the cells of patients with cystinosis has been seen as evidence that protein cystine residues are not reduced. The data are reconcilable and fully in harmony if it is postulated that cysteine from the cytoplasm ...
متن کاملDrug Profile
Cystinosis is a rare autosomal recessive disorder caused by mutations in the CTNS gene located on the short arm of chromosome 17 [1,2]. The CTNS gene encodes a lysosomal cystine transporter protein, cystinosin, which helps facilitate the efflux of cystine from lysosomes. The accumulation of the disulfide amino acid cystine within cellular lysosomes leads to the formation of cystine crystals in ...
متن کاملFolate pro-drug of cystamine as an enhanced treatment for nephropathic cystinosis.
Nephropathic cystinosis is a rare autosomal recessive disease characterised by raised intracellular levels of the amino acid, cystine. If untreated, the disease, progressively deteriorates towards end stage renal disease (ESRD) at the end of the first decade. The disease is caused by a defect in the lysosomal transport mechanism for cystine. The treatment of choice is the aminothiol cysteamine ...
متن کاملA randomized clinical trial of topical cysteamine disulfide (cystamine) versus free thiol (cysteamine) in the treatment of corneal cystine crystals in cystinosis.
In nephropathic cystinosis, corneal cystine crystals cause severe photophobia and corneal erosions. Topical cysteamine dissolves these crystals, but cannot be marketed because it rapidly oxidizes to the disulfide form, cystamine, at room temperature. Since cystamine itself could be used commercially, we compared the efficacy of cystamine and cysteamine with respect to cystine crystal dissolutio...
